Anti-cariogenic Activity of a Bacteriocin Produced by Lactococcus sp. HY 449

Abstract
The purpose of this study was to evaluate bacteriocin activity against cariogenic bacteria including Streptococcus mutans. The spot-on-lawn method was used to determine the antimicrobial activity against indicator strains. Abacteriocin produced by Lactococcus sp. HY 559 inhibited the growth of S. mutans 3065, S. mutans MT 8148, S. mutans CNU, S. sorbrinus ATCC 27607, S. mitis KCTC 3556, and S. vestibularis KCTC 3650. The bacteriocin was partially purified by ammonium sulfate precipitaion and hydrophobic interaction chromatography (HIC). Using chromatography with octyl-sepharose CL-4B, higher molecular weight and undesirable molecules were eluted with ammonium sulfate and H_2O, while the bacteriocin was eluted with ethanol. Purified bacteriocin was added to S. mutans MT 8148 cells during logarithmic growth. After 5 hr of incubation, the optical density (O.D. 660 nm) of cells without bacteriocin increased. Whereas the O.D. of cell suspensions decreased in the presence of bacteriocin, indicationg that bacteriocin appears to cause cell lysis of tested strains. The addition of bacteriocin statistically showed decrease of plaque formation (p<0.05). The mean weight of plaque in the culture of S. mutans MT 8148 exhibited 82.8 mg/20 mL of M17-5% sucrose broth, whereas the mean weight of plaque was 11.8 mg/20mL of M17.5% sucrose broth in the presence of 100 AU/mL bacteriocin. These results suggest that bacteriocin produced by Lactococcus sp. HY 449 is potentially useful in controlling cariogenic bacteria.
